產品介紹 | 組蛋白翻譯后修飾(PTMs)是表觀遺傳學調控染色質結構的關鍵機制,被稱為“組蛋白密碼”。組蛋白上的翻譯后修飾包括乙?;?,甲基化,磷酸化和近年發(fā)現(xiàn)的一些新型酰化修飾。這些組蛋白修飾直接影響染色質和轉錄因子或其他表觀調控子的結合,改變基因組的穩(wěn)定性和基因轉錄等。組蛋白甲基化通常發(fā)生在核心組蛋白的賴氨酸和精氨酸殘基。組蛋白甲基化既可以促進也可以抑制基因轉錄,這取決于組蛋白甲基化是發(fā)生在賴氨酸還是發(fā)生在精氨酸上以及甲基化基團的個數(shù)(賴氨酸可以發(fā)生一、二、三甲基化,精氨酸可發(fā)生單、對稱與非對稱二甲化)。組蛋白賴氨酸甲基化通常發(fā)生在組蛋白H3第4,9,27,36,79和組蛋白H4的第20位賴氨酸上;精氨酸甲基化通常發(fā)生在組蛋白H3的第2,8,17,26和組蛋白H4第3位精氨酸上。蛋白甲基化酶(HMT)和組蛋白去甲基化酶(HDM)是甲基化修飾主要的調控因子。 |
